Introduction

Avegene 96-Well SEQ Dye Clean Up Kit is designed for high-throughput

purification of DNA sequencing reactions. Ultra filtration (UF) membrane is

used to separate extremely small particles and dissolved molecules based

on their size. Salts and unincorporated dye terminators could be effectively

removed from reaction mixture without alcohol precipitation or gel filtration.

The kits are compatible with ABI Prism BigDye, Beckman CEQ or

Amersham DYEnamic ET Dye Terminator chemistres and can be used for

manual filtration or with robotic handing systems. The entire procedure can

be completed in 20 minutes.

Quality Control

The quality of 96-Well SEQ Dye Clean Up Kit is tested on a lot-to-lot basis.

The Kits are tested by deterrmining the presence of contamination signals

in sequencing electropherogram.

Additional Required

2. Loading Buffer

Caution

Buffer contains irritant agent. During operation, always wear a lab coat,

disposable gloves, and protective goggles.

 

Vacuum Protocol

1. Add 2X volume of Dye Wash Buffer to sequencing reaction

product. (If your product volume less then 20 µl, adjust final

volume to 20 µl with water). Mix gently by pipetting several times.

2. Place the Dye Remove Plate on the vacuum manifold.

3. Transfer diluted sample from Step 1 into the bottom of each well.

4. Set the vacuum to 25" Hg and turn on the vacuum pressure for 5-10

minutes until each well was empty.

5. Turn off the vacuum.

6. Add 100 µl Dye Wash buffer to each well on the plate.

7. Turn on the vacuum pressure for 5-10 minutes or until each well was

empty.

8. Turn off the vacuum and remove the plate from the manifold.

9. Press the plate on an absorbent material (tissue paper) to blot out the

excess liquid from the bottom of the plate.

10. Place the Dye Remove Plate on the vacuum manifold and turn on the

vacuum pressure for 5 minutes.

11. Add 20-30 µl of sequencing loading buffer or deionized water

into central of the membrane. Pipet up and down 5-10 times to elute

the sequencing product.

12. Transfer eluted products to a new 96 well 0.35ml collection plate.

Centrifuge Protocol

1. Add 2X volume of Dye Wash Buffer to sequencing reaction

product. (If your product volume less then 20 µl, adjust final

volume to 20 µl with water). Mix gently by pipetting several times.

2. Place the Dye Remove Plate on the waste collection plate( not

provide ).

3. Transfer diluted sample from Step 1 into the bottom of each well.

4. Centrifuge for 5 min at 3,500 rpm in a centrifuge.

5. Add 100 µl Dye Wash buffer to each well on the plate.

6. Centrifuge for 20 min at 3,500 rpm in a centrifuge.

7. Add 20-30 µl of sequencing loading buffer or deionized water into

central of the membrane. Pipet up and down 5-10 times to elute the

sequencing product.

8. Transfer eluted products to a new 96 well 0.35ml collection plate.
